The dopaminergic hypothesis of migraine, first suggested by clinical and pharmacological evidence, has recently received renewed interest following the demonstration that specific polymorphisms of the dopamine D2 receptor gene seem to be involved in the susceptibility psychiatric comorbidity and clinical expression of migraine. In the following paper we critically review clinical, pharmacological, biochemical, genetic and neuroimaging data supporting the involvement of dopamine in migraine, emphasizing certainties and limitations of this hypothesis.
